Pure Protein has developed a peptide binding assay based on soluble HLA and fluorescent polarization detection. This assay enables high throughput screening and quantification of peptide-HLA binding affinity in a robust, repeatable system.
Fluorescence polarization (FP) allows the direct measurement of the ratio between free and bound labeled ligand in solution without any separation steps. The technique of FP is based on the principal that if a fluorescent-labeled peptide binds to the sHLA molecule of higher molecular weight, polarization values will increase due to the slower molecular rotation of the bound probe.
To elaborate the binding of a peptide to MHC class I molecules, a reference fluorescent- labeled peptide is incubated with activated sHLA in the presence or absence of a competitor and peptide/MHC interaction is monitored over time.
