PolyScreen – Accurate and Reliable

Competition binding assay methodologies have become exceedingly popular for assessing the ability of synthetically defined peptide epitopes to associate with specific HLA class I complexes. This has been accomplished by comparing the relative affinities of multiple peptide ligands for the same HLA receptor. PolyTest will accurately determine inhibitory concentrations (IC50), representative of the affinity of the ligand-recognition site by incubating sHLA with a labeled reference peptide in the presence of different concentrations of competitor.

The PolyTest approach is used to validate screening results of parental epitopes by determining their accurate affinity in order to judge their immunogenic potential. In addition, PolyTest is also useful to evaluate peptides, which were identified by other epitope screening methodologies. Furthermore, PolyTest can be a major tool in comparative studies by comparing a native, unchanged peptide with a peptide whose sequence has been altered in order to overcome immune tolerance in cancer vaccine design or in determination of escape mutants in viral variance studies.
